Product overview

Description & Application

OXF-232 is a triazole-linked tazobactam–dipeptide conjugate investigated as a DegP-mediated TEM β-lactamase degrader. In a purified-protein assay, it promoted TEM-116 degradation with a DC50 of 5.8 ± 0.14 µM after 6 h. It is a research candidate for bacterial targeted protein degradation and β-lactam antibiotic-adjuvant studies.

Antibiotic-adjuvant activityImproved bacterial growth inhibition versus tazobactam at 1 mg/mL piperacillin; poorer performance at lower piperacillin concentrationsE. coli harbouring pK18-TEM-1; kinetic growth assay; piperacillin 0.1, 0.33 or 1 mg/mLMain Fig. 3A–C, pp. 5–6; SI Table S1

Evidence limits

  • Biochemical degradation and bacterial growth inhibition are different endpoints.
  • At 100 µM for 7 h without piperacillin, OXF-232 did not show detectable TEM-1 degradation in E. coli; enhanced cellular degradation must not be assumed for this series.
  • At lower piperacillin concentrations (0.1 and 0.33 mg/mL), tazobactam performed better; at 1 mg/mL piperacillin, the degrader series showed improved growth inhibition.
  • The term “in vivo” in the bacterial assays refers to living bacteria, not animal infection efficacy.
  • No DC stock, purity, lead time or clinical efficacy is established by the literature.

These values are reported in the cited research and depend on the experimental conditions. DC Chemicals supplies research compounds; literature results are not guaranteed product performance.
