Biological Activity
PTC299 (Emvododstat) is a DHODH inhibitor for pyrimidine metabolism and hematological-cancer research. PTC299 inhibited DHODH more strongly in the mitochondrial context, linking activity to the native enzyme environment.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| DHODH mechanism | Purified enzyme and isolated-mitochondria comparisons. [1] | PTC299 inhibited DHODH more strongly in the mitochondrial context, linking activity to the native enzyme environment. |
| Pyrimidine dependence | Hematologic cancer-cell models. [1] | Lower uridine-nucleotide availability inhibited proliferation; reduced salvage capacity helped explain sensitivity. |
| VEGFA interpretation | Phenotypic VEGFA-translation response. [1] | Suppression of VEGFA production was linked to pyrimidine depletion rather than establishing direct VEGF-receptor inhibition. |
In Vivo
No compound-specific animal efficacy result is established by the sources summarized here.
Clinical / Human Evidence
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Human target engagement | Archived serum from PTC299-treated participants. [1] | Increased dihydroorotate, the DHODH substrate, supported target engagement; it is not proof of clinical antitumor efficacy. |
Interpretation
PTC299 should not be described only as a direct VEGF inhibitor; altered VEGF production can follow pyrimidine depletion.
