Biological Activity
JP-153 is a focal-adhesion-signaling modulator for VEGF-dependent retinal endothelial-cell and angiogenesis research. JP-153 disrupted FAK–paxillin interactions and reduced paxillin Tyr118 and Akt Ser473 phosphorylation.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Focal-adhesion signaling | Human retinal endothelial cells stimulated with VEGF. [1] | JP-153 disrupted FAK–paxillin interactions and reduced paxillin Tyr118 and Akt Ser473 phosphorylation. |
| Functional readouts | Retinal endothelial migration and proliferation assays. [1] | Both responses decreased; initial Src and FAK activation were not prevented, distinguishing complex modulation from simple Src/FAK inhibition. |
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Retinal angiogenesis | Mouse oxygen-induced retinopathy; topical JP-153 microemulsion. [1] | Pathological neovascular tuft formation decreased in a dose-dependent manner. |
| Important accompanying effect | Same retinal model. [1] | Avascular area increased. Reduced tuft formation should not be described as uniform restoration of normal retinal vasculature. |
Interpretation
Formulated ocular efficacy cannot be directly assigned to unformulated JP-153 or a different route of administration.
