Biological Activity
AG1 is a small-molecule G6PD activator for enzyme oligomerization, NADPH metabolism and redox research. AG1 increased activity by approximately 20% and 60%, respectively, in the reported characterization.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Enzyme activation | Wild-type and Canton-mutant G6PD biochemical assays. [1] | AG1 increased activity by approximately 20% and 60%, respectively, in the reported characterization. |
| Mechanism | G6PD oligomerization, docking and mutational analyses. [1] | Noncovalent activation was consistent with stabilization of the dimer interface near structural NADP-binding sites. |
| Cell and erythrocyte response | Original discovery study; cellular and human erythrocyte assays. [2] | AG1 reduced oxidative-stress readouts, including stress associated with chloroquine or diamide in erythrocytes. |
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Zebrafish evidence | Original discovery study. [2] | Reduced oxidative stress was also reported in zebrafish. Mammalian clinical efficacy is not established by these findings. |
Interpretation
The cited work supports biochemical activation; clinical treatment of G6PD deficiency was not established.
