Biological Activity
Elafibranor (GFT505) is a dual PPARα/δ agonist used in hepatic lipid metabolism, inflammation and fibrosis research. Triglyceride content and Oil Red O staining evaluated the lipid response. The retrieved methods do not specify the subsequent GFT505 incubation duration.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Cellular lipid accumulation | LO2 hepatic cells loaded with oleate/palmitate (0.8 mM total, 2:1) for 24 h, then exposed to GFT505 7.5 or 15 µM. [1], Methods | Triglyceride content and Oil Red O staining evaluated the lipid response. The retrieved methods do not specify the subsequent GFT505 incubation duration. |
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Diet-induced liver disease | Male C57BL/6J mice on CDAHFD; GFT505 3, 10 or 30 mg/kg from weeks 5–12; n=10 per group. [1] | The paper assessed established diet-induced steatosis, inflammation and fibrosis. Route and dosing frequency were not explicit in the reviewed methods. |
| Histology and biomarkers | Liver histology, α-SMA/collagen-I and serum biochemistry. [1] | Elafibranor improved steatosis, inflammatory changes and fibrosis-related measurements. |
| Transcriptomic response | Treated mouse liver RNA sequencing. [1] | Broad changes in metabolic and inflammatory pathways accompanied histological responses; pathway enrichment is not a direct binding assay. |
Interpretation
These model-specific responses do not establish equivalent effects in all steatohepatitis models.
