Biological Activity
MK-8722 is a direct allosteric activator of mammalian AMPK complexes for metabolic signaling and glucose-homeostasis research. MK-8722 acted as a direct allosteric pan-AMPK activator.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| AMPK activation | Biochemical characterization of all 12 mammalian AMPK complexes. [1] | MK-8722 acted as a direct allosteric pan-AMPK activator. |
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Metabolic response | Rodent and rhesus-monkey studies, including diabetic monkeys. [1] | Stimulated insulin-independent skeletal-muscle glucose uptake and glycogen synthesis, with durable glycemic improvement. |
| Glycemic tolerability | Reported cross-species experiments. [1] | No evidence of hypoglycemia was observed under the studied regimens. |
| Cardiac limitation | Cardiac assessments after systemic activation. [1] | Cardiac hypertrophy and increased cardiac glycogen accompanied metabolic effects, although apparent functional sequelae were not detected in the reported experiments. |
Interpretation
The cardiac findings are a material limitation; improved glucose control does not imply a favorable overall safety profile.
