Biological Activity
CB-002 (CB002) is a p53-pathway-restoring research compound studied in colorectal cancer cells and apoptosis signaling. CB-002 increased NOXA, DR5 and p21-associated responses and promoted cell death.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| p53-pathway response | Mutant-p53 colorectal cancer models. [1] | CB-002 increased NOXA, DR5 and p21-associated responses and promoted cell death. |
| Functional dependence | NOXA knockdown and PARP-cleavage assays. [1] | NOXA depletion reduced cell-death and PARP-cleavage responses, supporting a mechanistic role for this apoptotic mediator. |
| Mutant-p53 stability | Models including RXF393; proteasome-inhibitor comparison. [1] | The study linked mutant-p53 destabilization to proteasomal degradation in responsive cells. |
| Normal-cell comparison | Normal fibroblasts under the reported experimental conditions. [1] | The study reported a differential response relative to tumor cells; no universal normal-tissue safety margin can be inferred. |
In Vivo
No compound-specific animal efficacy result is established by the sources summarized here.
Interpretation
Pathway restoration does not establish direct binding to, or universal repair of, every mutant p53 protein.
