Biological Activity
Praliciguat is an sGC stimulator for nitric-oxide/cGMP signaling, renal inflammation and fibrosis research. Praliciguat lowered proinflammatory gene expression and MCP-1 secretion.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Inflammatory response | Human primary renal proximal tubular epithelial cells challenged with TNFα. [1] | Praliciguat lowered proinflammatory gene expression and MCP-1 secretion. |
| Profibrotic signaling | TGFβ-challenged renal epithelial cells. [1] | Reduced SMAD3 phosphorylation, apoptosis and acquisition of a mesenchyme-like phenotype. |
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Diabetic nephropathy | Obese ZSF1 rats; praliciguat alone or with enalapril. [1] | Both praliciguat regimens attenuated proteinuria; monotherapy did not alter hemodynamics. |
| Comparator and renal markers | Enalapril and renal tissue endpoints. [1] | Enalapril alone lowered blood pressure without reducing proteinuria. Praliciguat lowered renal inflammation/fibrosis-associated gene expression. |
Interpretation
Cellular and animal findings are complementary; they do not establish a clinical renal benefit.
