Biological Activity
DO-264 (DO264) is an ABHD12 inhibitor for lysophosphatidylserine metabolism and immune-lipid signaling research. DO-264 inhibited this lipid hydrolase and increased lyso-PS in primary human macrophages.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Target function | ABHD12-dependent lysophosphatidylserine metabolism. [1] | DO-264 inhibited this lipid hydrolase and increased lyso-PS in primary human macrophages. |
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Brain pharmacodynamics | Adult mice treated with DO-264. [1] | Brain lyso-PS increased, supporting in-vivo inhibition of ABHD12. |
| Auditory phenotype | Pharmacological treatment compared with genetic ABHD12 deletion. [1] | Adult drug-treated mice showed minimal auditory disturbance, unlike the knockout phenotype. |
| Inflammatory limitation | Immune-challenge experiments. [1] | ABHD12 inhibition was associated with heightened inflammation and severe lung pathology. These results preclude describing the compound as generally innocuous. |
Interpretation
Pharmacological inhibition does not reproduce every phenotype of genetic deletion; immune and neurological effects require separate evaluation.
