S1PR2-associated signaling · Literature evidence
Biological Activity
JTE-013 is an S1PR2 antagonist used in sphingosine-1-phosphate signaling, mitochondrial dynamics and fibrosis research. Reduced RHOA/TAZ and CTGF/CYR61 signaling; phosphorylated YAP and Lats1 increased.

Epithelial signaling
MLE-12 cells; JTE-013 2 µM for 2 h before S1P 3 µM for 24 h. [1], Methods and Figs. 4–6
Reduced RHOA/TAZ and CTGF/CYR61 signaling; phosphorylated YAP and Lats1 increased.
Fibrosis and mitochondria
S1P-challenged MLE-12 cells. [1]
Lower α-SMA, COL1A1 and MMP-9 accompanied changes in Drp1-associated mitochondrial dynamics and reduced oxidative stress.

Exposure and model
Bleomycin lung-fibrosis mouse model; JTE-013 8 mg/kg intraperitoneally on days 1, 3 and 5; n=10 per group. [1], Methods
Lung inflammation and fibrosis were assessed on days 7 and 28.
Lung outcomes
Histology, collagen staining and bronchoalveolar inflammatory markers. [1], Figs. 1–3
JTE-013 reduced inflammatory infiltration and fibrosis-related changes. Verteporfin co-treatment reversed several measured effects.
Interpretation
Knockdown-specific numerical outcomes are not assigned to JTE-013 treatment.
