Biological Activity

MC3343 is a nonnucleoside DNMT inhibitor for epigenetic reprogramming, osteoblastic differentiation and osteosarcoma research. MC3343 arrested cells in G1 or G2/M and re-expressed genes associated with osteoblast differentiation.

In Vitro

EndpointModel / conditions / sourceReported result
Proliferation and differentiationHuman osteosarcoma models. [1]MC3343 arrested cells in G1 or G2/M and re-expressed genes associated with osteoblast differentiation.
Comparison with 5-azadCOsteosarcoma cells; DNMT-inhibitor comparison. [1]Antiproliferative effects were similar, but the differentiation response differed from the nucleoside inhibitor.
Chemotherapy combinationsMC3343 with doxorubicin or cisplatin. [1]Combination effects included increased stable doxorubicin–DNA association, sustained DNA damage and greater cell death.

In Vivo

EndpointModel / conditions / sourceReported result
Patient-derived modelOsteosarcoma PDX. [1]A mature osteoblast phenotype and sustained cytostatic response were reported in vivo. Exact exposure conditions are not available in the verified abstract.

Interpretation

Differentiation and cytotoxicity are separate outcomes; combination findings are not monotherapy response rates.
