Biological Activity
KML-29 is a selective MAGL inhibitor used to investigate endocannabinoid metabolism, inflammatory pain and neuropathic pain. Reduced paw edema and mechanical allodynia.
In Vitro
No separate compound-specific cell or biochemical result is available from the sources summarized here.
In Vivo
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Inflammatory pain | Mouse carrageenan model; KML-29 20 or 40 mg/kg intraperitoneally; 2 h pretreatment. [1] | Reduced paw edema and mechanical allodynia. |
| Brain lipid response | Acute KML-29 treatment; brain lipid measurements. [1] | 2-Arachidonoylglycerol increased at 2–4 h and returned toward baseline by 24 h; arachidonic acid decreased. |
| Neuropathic pain | Chronic constriction injury; 40 mg/kg intraperitoneally. [1] | Reduced cold allodynia at 3–6 h and mechanical allodynia, with a peak mechanical response at 3 h. |
| Repeated-dose limitation | 40 mg/kg daily for 6 days. [1] | The anti-allodynic response was lost with repeated treatment, accompanied by CB1 receptor desensitization. |
| Behavioral tolerability | Selected acute behavioral tests. [1] | No catalepsy, hypothermia or hypomotility was detected under the tested conditions; this is not a general absence-of-toxicity claim. |
Interpretation
The absence of selected cannabimimetic effects in this study is not a general safety guarantee.
