Biological Activity
Y-27632 dihydrochloride is a ROCK inhibitor for cell-survival and cytoskeletal research, including dissociated human pluripotent stem-cell cultures. Y-27632 reduced dissociation-induced apoptosis and increased cloning efficiency from approximately 1% to approximately 27%.
In Vitro
| Endpoint | Model / conditions / source | Reported result |
|---|---|---|
| Dissociated stem-cell survival | Human embryonic stem cells after dissociation. [1], abstract | Y-27632 reduced dissociation-induced apoptosis and increased cloning efficiency from approximately 1% to approximately 27%. |
| Clonal expansion | Human embryonic stem cells following gene transfer. [1] | Improved survival facilitated subcloning from single cells. |
| Suspension culture | Serum-free floating embryoid-body-like aggregate culture. [1] | Dissociated cells survived and formed floating aggregates after Y-27632 treatment. |
| Differentiation outcome | Human embryonic stem-cell aggregates. [1] | Surviving cells differentiated into Bf1-positive cortical and basal telencephalic progenitors. The verified abstract does not specify the complete exposure schedule. |
In Vivo
No compound-specific animal efficacy result is established by the sources summarized here.
Interpretation
The reported improvement is specific to the study system; it is not a universal survival percentage for every cell line.
