In TNBS-induced colitis mice, rectally administered Fluc mRNA MP2 LNPs gave approximately 15-fold higher colonic luminescence than the study's SM102 formulation at 6 hours (0.5 mg/kg mRNA; n = 3 mice per group; Fig. 1g).

Reported study results
MP2 was among the five three-tail steroid-derived formulations with more than 30-fold higher Fluc mRNA expression than the study's SM102 formulation in Caco-2 cells (0.5 µg/mL mRNA; 18 hours; n = 4 wells per group; Fig. 1f).
IL-35 mRNA-loaded MP2 LNPs improved clinical and colonic inflammation measures in TNBS acute and IL-10 knockout chronic mouse colitis models. The IL-10 knockout study reported disease activity index 0.38 for MP2-IL-35 versus 3.75 for SM102-IL-35 at week 18 (1 mg/kg mRNA rectally each week; Fig. 4).
MP2 LNPs delivered GFP mRNA to immune, epithelial and endothelial cells in human colonic biopsies. IL-35 mRNA-loaded MP2 LNPs were also studied in IBD patient biopsies for cytokine and T-cell readouts (Fig. 5).
