Reported measurements and study results

Selection

BP-200-C12, BP-197-C12 and BP-488-C10 were top in the BJ fibroblast luciferase-mRNA screen. BP-200-C12 was selected for subsequent studies (Figure 1c–e).

Hydroxyapatite adsorption

After 120 min, fluorescently labeled MiTEX showed 80% HA binding versus 10% for the C12-200 LNP comparator (Figure 2c,d). The lipid and whole LNP are distinct.

Solid-phase RNA delivery

Following 2,000 ng input mRNA and substrate washout, about 200 ng mRNA remained on HA in the MiTEX condition versus less than 50 ng in the control (Figure 3b). Cre-mRNA-loaded HA labeled approximately 80% of tdROSA mMSCs versus 17.92% with the control (Figure 3e,f).

Bone graft and gingival assays

Cre-mRNA-loaded HA/TCP with donor mMSCs was implanted subcutaneously in mice and assessed at 60 days (Figure 4). Separately, STAT3-siRNA MiTEX reduced pSTAT3 and IL-8 in HA-associated gingival fibroblast experiments; the human explant IL-6 readout was significant while IL-8 was not (Figure 5).

The formula and molecular weight are calculated from the fully protonated phosphonic-acid drawing; the paper does not report a measured mass for this final compound. doi:10.1016/j.biomaterials.2026.124680
