79.6 nm Z-average diameter, PDI 0.134, 99.40% mRNA encapsulation and zeta potential +1.13 mV (Supplementary Table S1). These characterize the formulated LNP, not the neat lipid.

Reported measurements and biological results
With the optimized lipid-6 protocol, >90% eGFP-positive dendritic and NK cells and >97% eGFP-positive tumor-infiltrating lymphocytes were reported. Activated primary T cells exceeded 95%; nonactivated T cells exceeded 80% (Figure 4; three donors).
Approximately 90% CAR-positive primary human T cells 24 h after transfection with lipid-6 LNPs carrying MUC1-CAR mRNA (Figure 5C). The engineered cells showed greater in vitro killing of MUC1-positive MDA-MB-468 and SKOV3 cells than controls in the reported co-culture assays (Figures 6 and 7; three donors). This is not an in vivo CAR-T efficacy result.
After 0.5 mg/kg firefly luciferase mRNA, lipid-6 LNPs produced higher bioluminescence than SM-102 and ALC-0315 comparators at 6 h for both intravenous and intramuscular administration (Figure 2; n = 3 per group). The mRNA cargo and endpoint differ from the CAR-T assay.
