Formulation & Performance

Study-specific compositions, preparation conditions and experimental outcomes for TD5.

Evidence coverage

Publisher abstract rechecked. Previously extracted composition and characterization are retained separately; full-text/SI dose-and-time verification remains incomplete in this revision.

1. Formulations and component amounts

Reported composition and calculated mass are separate. Molecular-weight inputs use the current catalog values; DMG-PEG2000 is fixed initially at 2507. Edit inputs only when using a verified alternative material.

Calculated total: 100.0000 mg lipid. RNA, solvent and process loss are excluded.

How component weights are calculated

For total lipid mass M (mg), molar parts rᵢ and molecular weight MWᵢ: component mass = M × rᵢ × MWᵢ / Σ(r × MW). Component amount (µmol) = component mass (mg) × 1000 / MWᵢ. Molar parts are normalized only for the displayed percentage. The calculation does not infer N/P or RNA dose.

2. Preparation and experimental setup

ParameterReported procedure / limitationSource
AdministrationIntrathecal delivery is the route supported by the publication abstract. It is not evidence for systemic crossing of the blood–brain barrier.Publisher abstract
Recipe / N/PPrior record gives the four-component composition above. N/P, phase ratio, mixing flow and purification have not been re-verified from complete Methods.Prior extraction; primary protocol pending
Dose mappingThe earlier record lists 0.135 mg/kg for a mouse intrathecal experiment. That dose has not been independently mapped to every GFP, Cre and CRISPR endpoint in this revision. Not propagated into all result rows

4. Performance by experiment

Experiment and doseReadout and comparatorResult and source
GFP expression

Mouse brain; TD5 BLNP–GFP mRNA; single intrathecal injection

Readout: Dose/time not specified in the accessible abstract

Comparator: MC3 LNP comparison described by the authors

Reported GFP-positive neurons 29.6% and astrocytes 38.1%. These are cell-population percentages, not brain fraction of injected dose.Publisher abstract
Cre reporter recombination

Ai14 mice; TD5 BLNP–Cre mRNA; intrathecal

Readout: Dose/time not specified in the accessible abstract

Comparator: Study reporter controls

Approximately 30% of neurons and 40% of astrocytes showed tdTomato expression across major brain regions. These values are not Cas9 editing efficiencies.Publisher abstract
CRISPR delivery

TD5 BLNP carrying Cas9 mRNA/sgRNA; mouse brain

Readout: Dose/time and numerical editing rate require full text

Comparator: Study editing controls

The abstract reports effective brain genome editing; no exact percentage is supplied here.Publisher abstract

5. Interpretation and unresolved details

  • Full paper and SI are still needed for dose, volume, injection site, readout time, replicate count, statistics and exact preparation details.
  • The mass worksheet is based on the earlier extracted composition and current catalog values; it is not an independently re-verified administration protocol.
  • Intrathecal delivery and systemic brain targeting are different experimental claims.
  • GFP expression, Cre recombination and Cas9 editing must remain separate endpoints.

Results apply to the stated LNP composition, cargo, process, dose, route and model. LNP experimental measurements are not batch-release specifications for the isolated lipid.
