Formulation & Performance

Study-specific compositions, preparation conditions and experimental outcomes for Lipid 43.

Evidence coverage

Patent PDF reviewed, including the TMF30 composition, particle-size table, administration method and tissue-luminescence table.

1. Formulations and component amounts

Reported composition and calculated mass are separate. Molecular-weight inputs use the current catalog values; DMG-PEG2000 is fixed initially at 2507. Edit inputs only when using a verified alternative material.

Calculated total: 100.0000 mg lipid. RNA, solvent and process loss are excluded.

How component weights are calculated

For total lipid mass M (mg), molar parts rᵢ and molecular weight MWᵢ: component mass = M × rᵢ × MWᵢ / Σ(r × MW). Component amount (µmol) = component mass (mg) × 1000 / MWᵢ. Molar parts are normalized only for the displayed percentage. The calculation does not infer N/P or RNA dose.

2. Preparation and experimental setup

ParameterReported procedure / limitationSource
Formulation identityTMF30 contains compound 43; it is not a generic formulation for every lipid in the patent.Table 3
Preparation cross-referenceThe patent states TMF17–TMF53 follow the TMF2 preparation with the Table 3 component substitutions. Exact TMF2 mixing/buffer parameters are not reproduced in this revision.Paragraph [0511], printed p.129
RNA ratio / N/PNot transcribed from a verified preparation passage; no inferred N/P is supplied.Preparation cross-reference requires completion
Animal reporter assayFemale ICR mice, 6–8 weeks; retro-orbital intravenous administration, 0.5 mg/kg RNA; luciferin 150 mg/kg before imaging.Paragraph [0517], printed p.132

4. Performance by experiment

Experiment and doseReadout and comparatorResult and source
Spleen reporter expression

ICR mice; luciferase RNA; intravenous 0.5 mg/kg

Readout: 4 h after dosing

Comparator: TMF17, DODAP-based formulation with the same listed helper ratios

TMF30 spleen flux 1.31 × 10⁹ p/s; TMF17 1.45 × 10⁶ p/s. These are tissue expression signals, not percentages of injected particles.Table 5, printed p.134; n/statistical test not stated in the transcribed table
Liver reporter expression

Same TMF30 reporter experiment

Readout: 4 h

Comparator: TMF17 liver flux 1.69 × 10⁶ p/s

TMF30 liver flux 3.56 × 10⁷ p/s. Higher spleen signal does not imply absence of liver delivery.Table 5; paragraph [0517]
Spleen:liver selectivity

Same assay; compare individual formulations

Readout: 4 h

Comparator: TMF29 (compound 38): spleen 5.73 × 10⁸, liver 8.48 × 10⁶ p/s; reported ratio 69.5

TMF30 reported ratio is 38.8; dividing its displayed mean fluxes gives 36.80 (calculated). TMF30 has greater absolute spleen flux than TMF29 but a lower reported spleen:liver ratio.Table 5; calculated ratio is identified separately

5. Interpretation and unresolved details

  • The patent-reported 38.8 ratio is retained as reported; the displayed means do not reproduce it exactly. The statistic underlying that ratio is not specified here.
  • No universal “best lipid” ranking follows from these data: absolute spleen expression and spleen:liver selectivity rank formulations differently.
  • This is patent-stage mouse reporter evidence. CAR expression, human performance and therapeutic efficacy are not established by the cited TMF30 table.

Results apply to the stated LNP composition, cargo, process, dose, route and model. LNP experimental measurements are not batch-release specifications for the isolated lipid.
