Description:

A18B8C14 is an α-branched amine ionizable lipid developed using the photochemistry-based ionizable lipid library (PILL) platform. Its structure contains two secondary amines linked by a pentamethylene spacer and two ester-containing branched hydrophobic arms. A18B8C14 was identified as a lead for mRNA delivery; the publication reports higher in vivo expression with its optimized LNP formulation than with an MC3-based benchmark. The supporting information documents intravenous reporter-mRNA delivery, formulation screening, cellular uptake and endosomal colocalization, particle stability, and dose-dependent blood chemistry. It is intended for research on mRNA-LNP formulation and delivery.

StorageFollow the supplied batch label; protect from light and moisture. Compound-specific stock-solution shelf life is not established.

Handling:

Store the unopened product at −20 °C, protected from light and moisture, with the vial tightly sealed. The standard storage period is 1 year; follow the expiry or retest date on the supplied batch documentation. Allow the closed vial to reach room temperature before opening to avoid condensation.

We ship the pure lipid with ice packs, without ethanol. Prepare an ethanol stock solution when required. Gentle warming and brief sonication may assist dissolution; confirm complete dissolution at the intended concentration.

Prepare and use solutions on the same day wherever possible. If advance preparation is required, use a validated storage procedure and tightly sealed aliquots. A general solution shelf life is not assigned.

For small amounts of oily or viscous lipid, dissolve the entire pack and take volumetric aliquots. For example, dissolve 25 mg and adjust to a final volume of 1 mL to obtain 25 mg/mL; a 100 µL aliquot contains 2.5 mg.

A18B8C14-specific neat-material and stock-solution shelf life are not established by the LNP stability experiment. Follow the supplied batch label.

Formulation & Performance:

A18B8C14 / DSPC / cholesterol / DMG-PEG2000 is a tested combination in SI Fig. S3a at 50:10:38.5:1.5 mol%, with total lipid:mRNA 20:1 w/w. N/P is not reported. This screening formulation is not asserted as the final optimized or SORT formulation.
