Formulation & Performance

This edition organizes the previously reviewed, source-linked product record. It is not a new independent review of every paper or supporting file. Reported and calculated values retain their original labels.

Formulation & Performance

Study-specific compositions, preparation conditions and experimental outcomes for HI-62.

Evidence coverage

Limited source access: the existing product record has been reorganized. Full-text and supplementary-file re-verification remains pending; numerical results below are retained from that earlier record and are not newly independently verified.

1. Formulations and component amounts

Reported composition and calculated mass are separate. Molecular-weight inputs use the current catalog values; DMG-PEG2000 is fixed initially at 2507. Edit inputs only when using a verified alternative material.

No complete mass recipe is generated for this formulation while composition or identity-dependent inputs remain unresolved.

How component weights are calculated

For total lipid mass M (mg), molar parts rᵢ and molecular weight MWᵢ: component mass = M × rᵢ × MWᵢ / Σ(r × MW). Component amount (µmol) = component mass (mg) × 1000 / MWᵢ. Molar parts are normalized only for the displayed percentage. The calculation does not infer N/P or RNA dose.

Reporter expression

Mouse; FLuc mRNA; intravenous; 5 µg per mouse

Readout: 48 h

Comparator: SM-102 formulation

Earlier record reports >1,000-fold higher expression. Animal number, organ normalization and comparator recipe require primary-source confirmation.Earlier product record; figure not re-verified
Freeze–thaw response

RNA-loaded E21; six cycles

Readout: After six cycles; temperature protocol pending

Comparator: SM-102 formulation

Earlier record reports >95% encapsulation versus a comparator change from 96% to <90%. Expression after cycling is not established here.Earlier product record; source check pending
Tumor study

TC-1 mouse model; mRNA 10 µg per dose on days 0, 5 and 10

Readout: Response-assessment date pending

Comparator: Comparator details pending

Earlier record reports complete responses in 4/5 mice. Cargo identity, tumor schedule and response definition must be checked before protocol reuse.Earlier product record; source check pending

Mechanistic evidence

A thiourea-containing ionizable lipid is described in the earlier product record. Structure-based hypotheses are not substituted for measured uptake or endosomal-escape data.

5. Interpretation and unresolved details

  • The cited work is a preprint, not a peer-reviewed clinical study.
  • This guide does not provide a complete reproducible E21 recipe. A component-mass calculation is withheld because the composition is missing.
  • The full text and SI are required to resolve the assay conditions and source locations.

Results apply to the stated LNP composition, cargo, process, dose, route and model. LNP experimental measurements are not batch-release specifications for the isolated lipid.
