Literature formulation

AO16:SM-102:DSPC:cholesterol:DMG-PEG2000 = 2.50:50.00:10.00:38.50:1.50 reported molar ratio (sum 102.50; normalized values 2.44:48.78:9.76:37.56:1.46 mol%). Total lipid:mRNA = 20:1 by mass. N/P was not reported.

Reported screening conditions

Aqueous citrate buffer pH 4.0 was mixed with ethanol lipid phase at 3:1, vortexed 5 min, incubated 10 min, and dialyzed in PBS for at least 12 h. The source screened 250 ng mRNA per well and exposed formulations to 0.05–0.50 mM reactive species for 2 h.

Reported AO16 results

At 0.05 mM, the antioxidant score was 67% versus 57% for the commercial control LNP. At 0.50 mM, the score was 32% versus 37% for control. AO16 results are in vitro screening data; AO12-specific animal results from the same paper are not assigned to AO16.
