Optimized by changing CEBPA-51 concentration while holding lipid concentration constant; no single total-lipid:RNA mass ratio is stated in the reviewed text.
RNA phase: sodium acetate/sucrose pH 4.0. Immediate pH adjustment: NaCl/phosphate pH 9.0. Final exchange buffer: sucrose/phosphate pH 7.5.
CEBPA-51 small activating RNA (saRNA)
Intravenous infusion in the described product context; intravenous dosing in the rat cirrhosis example
1 mg/kg MTL-CEBPA in the delayed-treatment rat cirrhosis example
Male Sprague-Dawley rats with carbon-tetrachloride-induced cirrhosis
CEBPA-51 in sodium acetate/sucrose pH 4.0 and lipids in absolute ethanol at 55 C were combined by crossflow ethanol injection. The mixture was immediately diluted with NaCl/phosphate pH 9.0, extruded through 0.2 micrometer polycarbonate membrane, ultrafiltered, diafiltered with at least seven volumes of sucrose/phosphate pH 7.5, and sterile filtered.

Additional reported findings

pH-dependent loading

MOCHOL is described as positively charged in the acidic RNA-loading phase (pKa 6.5); raising pH after vesicle formation produces the charge-converted final system.

Storage degradation pathway

Under 25-40 C stress, MOCHOL content fell while cholesterol increased; the other lipid excipients and total saRNA did not show significant changes in the described test.
