AA-T3A-C12 is an anisamide-tethered ionizable lipidoid containing a T3A polyamine core and four C12 epoxide-derived tails. In a four-component siRNA LNP, the anisamide group promoted sigma-receptor-dependent uptake by activated fibroblasts. In CCl4-induced liver fibrosis in mice, intravenous AA-T3A-C12/siHSP47 LNP reduced HSP47 and collagen deposition relative to the PBS disease control. These literature results apply to the cited formulation and do not establish performance of the neat lipid alone.
