Component molecular weights and calculated amounts

Composition retained from the cited study below. Research planning aid only. Results cover lipid component weights and do not include RNA, buffer, solvent, process loss or an N/P ratio.

ComponentMolecular weightMolar partsNormalized mol%Amount (µmol)Weight (mg)

Calculated total: 100.0000 mg lipid.

Calculated catalog planning quantities; RNA, solvent and process loss are excluded. DMG-PEG2000 uses 2507. Component mass = total lipid mass × molar parts × molecular weight / sum(parts × molecular weight). This conversion does not infer N/P.

PBS pH 7.4 during collection and dilution; final dialysis to pH 7.3-7.4.
Firefly-luciferase mRNA or SARS-CoV-2 S2P mRNA, according to experiment

Reporter studies: 0.5 or 5 micrograms mRNA. Immunogenicity/challenge: 0.1, 0.25, 0.5 or 1 microgram S2P mRNA.
BALB/c mice and K18-hACE2 mice
Lipids in ethanol and mRNA in aqueous buffer were mixed with a Spark NanoAssemblr. The dispersion was ejected into PBS, diluted 1:1 with PBS and dialyzed with six buffer exchanges over 6 h using a 10 kDa device. Exact stream flow rates were not reported.

Additional reported findings

Liver off-target reporter signal

At 5 micrograms mRNA, liver signal was reported as approximately sixfold lower than MC3 (p=0.051).

Lung viral titers

C24 at 0.5 microgram eliminated detectable lung infection in the two animals assessed on day 5; MC3 did not completely eliminate infection even at 1 microgram.

RNA integrity

mRNA integrity was retained for at least 19 days at 4 C and declined more slowly for C24 than MC3 at higher storage temperatures.
