3.4
mRNA acetate buffer, pH 5.0 during encapsulation; buffer exchange/neutralization into PBS, pH 7.2; stored at 5 °C.
Modified hEPO mRNA containing miR126 and miR142 target sites; firefly luciferase mRNA in a confirmatory experiment.
Intravenous bolus in CD-1 mice; 60-minute intravenous infusion in male cynomolgus monkeys.
0.5 mg/kg hEPO mRNA in mouse expression studies; 0.1 mg/kg hEPO mRNA in cynomolgus-monkey infusion.
CD-1 mice (n=5 for primary expression comparisons); male cynomolgus monkeys (n=6 for pharmacodynamic comparison).
Modified ethanol-drop nanoprecipitation; lipid components in ethanol were combined with mRNA in acetate buffer.

Additional reported findings

PEG-lipid sensitivity

Lipid 29 maintained consistent particle size and low PDI across 0-3 mol% PEG-lipid, including 0 mol%; long-term stability at very low PEG was not determined.

· Article section 2.3, Fig. 6.
Proposed lipid–mRNA interactions

Molecular-dynamics simulations indicated squaramide 29 hydrogen-bond and pi-stacking interactions with exposed RNA nucleobases/sugars; this is a mechanistic hypothesis from simulations, not a standalone efficacy guarantee.
