Component molecular weights and calculated amounts

Composition retained from the cited study below. Research planning aid only. Results cover lipid component weights and do not include RNA, buffer, solvent, process loss or an N/P ratio.

Calculated total: 100.0000 mg lipid.

Calculated catalog planning quantities; RNA, solvent and process loss are excluded. DMG-PEG2000 uses 2507. Component mass = total lipid mass × molar parts × molecular weight / sum(parts × molecular weight). This conversion does not infer N/P.

FLuc mRNA at 1 mg/mL in 43-50 mM sodium acetate, pH 4; final dialysis against PBS to pH 7.3-7.4.
N1-methylpseudouridine-modified, CleanCap firefly-luciferase mRNA
HEK293 cell culture; intramuscular gastrocnemius injection or intravenous retro-orbital injection in separate mouse groups
In vitro 25, 50, 100 or 200 ng mRNA/well; in vivo 5 micrograms mRNA-LNP/mouse
HEK293 cells, 12,000 cells/well; 8-week-old female BALB/c mice
Lipids were dissolved in ethanol at 50 mM total lipid. A 16 microliter organic phase and 32 microliter aqueous phase were mixed in a Spark NanoAssemblr and ejected into 48 microliters PBS, then diluted with 96 microliters DPBS. LNPs were dialyzed against PBS for 4-6 h with 4-6 buffer exchanges and concentrated for in vivo use.
