Reported observation

Literature-reported Factor VII silencing Optimized 304O13 LNP EC50 0.01 mg/kg siRNA; after one 0.1 mg/kg injection, Factor VII remained suppressed for 18 days. Optimized PEG condition, anti-FVII siRNA IV · Non-targeting siRNA controls and non-optimized PEG conditions PDF p. 4, Fig. 3c-d.

Reported observation

Literature-reported Immune-cell delivery High CD45 silencing in CD11b+ monocyte/macrophage populations from peritoneal cavity and CD11c+ dendritic cells from spleen; peritoneal silencing up to approximately 90% and splenic silencing up to approximately 40% in the plotted results. 304O13 CD45-siRNA LNP, 3 days post IV injection · Non-targeting siRNA LNPs and other lipidoids PDF p. 4, Fig. 3e-f.

Reported observation

Literature-reported Biodistribution At 1 h, 304O13 Cy5.5-siRNA signal was primarily liver (42%) and spleen (24%), with 18% in kidneys; naked siRNA was 14% liver, 1% spleen and 71% kidneys. Cy5.5-siRNA, 1 mg/kg IV · Naked siRNA PDF p. 5, Fig. 4a-b.

Reported observation

Structure-function criteria Across the library, O13 tail, at least three tails and a tertiary amine were associated with potent Factor VII silencing; surface pKa of at least approximately 5.5 was also required for strong activity. These are library-level findings, not a 304O13-only causal experiment. · PDF pp. 6-7, Fig. 5 and Table 2.

Reported observation

Degradability 304O13 was hydrolyzed under acidic and basic conditions in the Methods; this demonstrates chemical degradability under the tested conditions and is not an in-vivo clearance half-life. · PDF p. 8, Lipidoid hydrolysis Methods.
