1. Handling

Keep the neat lipid tightly sealed, protected from light and stored at −20 °C. For a working stock, dissolve in a compatible solvent such as ethanol only after confirming solubility; prepare on the day of use where practical and keep aliquots at −20 °C for up to one month.

2. Literature starting point

For the reported intramuscular formulation, Lipid 331/DOPE/cholesterol/C14-PEG2000 was 35:16:46.5:2.5 mol%. For intranasal studies, the reported ratio was 30:39:30:1 with DOTAP replacing DOPE. The paper used microfluidic mixing at 3:1 aqueous:ethanol and an ionizable-lipid:mRNA mass ratio of 10:1.

3. Do not infer missing values

N/P was not reported. Do not convert the 10:1 mass ratio to N/P. The LNP component-mass calculator is intentionally disabled because the PEG-lipid identity/MW is not sufficiently specified. The product page separately provides a standalone Lipid 331 solution calculator using MW 978.52.

4. Literature physical data

65.4 ± 0.8 nm; PDI 0.2; zeta −6.2 ± 1.7 mV; EE 75.0 ± 8.9%; apparent pKa 7.53 ± 0.17. These values are specific to the cited study's materials, cargo, method and assay.
